This study aims to carry out antimicrobial activity test and fractionation of the compounds from the metabolite extract of Penicillium sp. (code FSKT 11) which is an endophytic fungi that was isolated from marine in Sekotong, Lombok. The process of the study was rejuvenation, seeding, fermentation, extraction, fractionation, spot monitoring, antifungal, antibacterial, and antimycobacterial activity test, and then determining the chromatogram profile of the active fraction using (thin layer chromatography) TLC and (high performance liquid chromatography) HPLC. The method used for fractionation of active compounds is (vacuum liquid chromatography) VLC. Meanwhile, the methods used for antimicrobial activity test include agar disc diffusion, microdilution, and microtetrazolium (MTT) assay. Microbes used in the activity tests include Candida albicans, Staphylococcus aureus, Escherichia coli, and Mycobacterium smegmatis. Based on the activity test, mycelium extract of Penicillium sp. is active against S. aureus and M. smegmatis with both of the MIC values are 200 ?g/mL and the MBC values are 400 ?g/mL and 200 ?g/mL respectively. The most active fraction is FB with the value of MIC and MBC against S. aureus are 100 ?g/mL and >800 ?g/mL. While the MIC and MBC values of FB against M. smegmatis are 200 ?g/mL. The results showed that the FB fraction had moderate antimicrobial activity and further investigation is possible to be carried out. text |
