Summary:Pegagan (Centella asiatica (L.) Urban) belongs to the Apiaceae family. It contains several secondary metabolite compounds such as asiaticoside and madecassoside that belongs to the pentacyclic saponin triterpenoid. These compounds have the prospect to be further developed into drug substances because of their pharmacological activities reported before and also used as raw material for cosmetic products, and food sources. However, there was a problem since pegagan can already be harvested from its natural habitat which has a long period to be growth and low secondary metabolite content. This study aimed to grow the optimization and production of asiaticoside and madecassoside through the callus culture. The optimization was carried out on MS solid medium, which was added with sucrose, agarose, growth hormone (NAA, 2,4 D, kinetin, tryptophan dan BAP), then sub-cultured into similar solid media that was stimulated with pectin 50; 100; and 200 mg/L also yeast extract 100 and 400 mg/L. Calluses are harvested in the fourth week, and subsequently must be dried and extracted for analysis using TLC and HPLC methods. Through TLC, callus with elicitor treatment did not have a clear spot of asiaticoside and madecassoside compound. Analysis by UPLC MRM MS/MS method showed that asiaticoside was detected at elicitation using pectin 100 mg/L; pectin 200 mg/L, and yeast extract 400 mg/L. Meanwhile, madecassoside were detected in all treatment except for the elicitation of 100 mg/L yeast extract. Elicitation by yeast extract and pectin did not give significant changes to the optimization of the content of asiaticoside and madesassoside compounds in pegagan.
