dangerous. Therefore, it is necessary to search for alternatives for better treatment. This study
aimed to determine the antihyperuricemia activity of the extracts and its fractions of C. sappan
and wedang uwuh. This research includes the preparation of extracts and fractions,
determination of total phenol and flavonoid content, monitoring of TLC profile, in vitro
xanthine oxidase inhibition test, and antihyperuricemia test in animal models of hyperuricemia
induced by potassium oxonate. Extraction of C. sappan content was carried out by reflux using
96% ethanol while wedang uwuh was by boiled. The results of the determination of total phenol
and flavanoid content revealed that the ethanolic extract of C. sappan contained 93.69 ± 1.83
mg GAE/g and 1.03 ± 0.01 mg QE/g of extract while wedang uwuh contained total phenols as
54 ± 0.004 mgGAE/g extract, but the total flavonoid content was not detected. In monitoring
the TLC profile, the ethanol extract, ethyl acetate fraction, and water fraction of C. sappan
contained phenols and flavonoids. The activity test results showed that the ethanol extract,
ethyl acetate fraction, water fraction, n-hexane fraction of C. sappan, and wedang uwuh
inhibited the xanthine oxidase enzyme with IC50 of 50.77, 36.97; 77.20; >200; >200 g/mL,
respectively. The in vivo test results showed that at 180 minutes, both doses of all fractions of
C. sappan and wedang uwuh were able to significantly inhibit the formation of uric acid
(p<0.05). Only the ethyl acetate fraction of C. sappan 50 mg/kg BW could inhibit the formation
of uric acid, which is comparable to allopurinol. Based on the inhibitory activity of xanthine
oxidase and the inhibition of uric acid formation comparable to allopurinol, it is supposed that
the ethyl acetate fraction of C. sappan acts as an antihyperuricemia by inhibiting xanthine
oxidase.
