Summary:C-phycocyanin (CPC) is one of the pigment-proteins contained in Spirulina platensis. A study stated that CPC can be used as a substrate for pancreatin protease (PP) activity assay. This study aims to obtain a high value CPC purification method based on purity and function as substrate for PP activity assay. Dry biomass of S. platensis was extracted using a combination of homogenization, sonication, centrifugation, and filtration. The extraction results were purified using Aqueous Two-Phase System (ATPS) –dialysis (method 1), the combination of ATPS –precipitation ammonium sulfate –dialysis (method 2), and the combination of precipitation ammonium sulfate –ATPS –dialysis (method 3). Isolation fraction was characterized using spectrophotometry UV-Vis, spectrofluorometry, determination of concentration and purity CPC, and application as a substrate in PP activity assay. The results of isolation produced 6 fractions, namely as purification fraction (F-PM1, F-PM2, and F- PM3) and dialysis fraction (FD1, FD2, and FD3). The highest purity CPC value was obtained from FD3, then the function as substrate on PP activity assay with highest reaction rate obtained from FD2. Based on this study, purification method for isolate CPC was recommended by using the method 3.
