Summary:One of the active pharmaceutical ingredients with the highest import value in Indonesia is amoxicillin. Conventional method of amoxicillin production was carried out by using 6- aminopenicillanic acid (6-APA) and D-4-hydroxyphenylglycine (D-4-HPG) by organic synthesis. To increase the process efficiency, the new method is catalyzed by enzyme, which requires D-4- hydroxyphenylglycine methyl ester (D-4-HPGME). The goal of this study was to obtain the optimum method of D-4-HPGME synthesis and its characterization data. The synthesis method was derived from patent CN103641729A, which uses one-pot reaction. D-4-HPG and methanol as reagent, while trimethylsilyl chloride as acid catalyst. The synthesis process was carried out by varying the number of precursors, as well as the temperature and reaction time. Isolation of the product was carried out by precipitation by adjusting the suspension mixture pH to 8.5. Followed by purification of the precipitate by recrystallization. The reaction at room temperature (120 hours) and at 60°C (5 hours) gave a yield of 73% and 71%, respectively. Characterization of the product by infrared spectroscopy and 1 H-NMR spectroscopy showed that the synthesized product was D-4-HPGME.
