The compounds derived from Gotu kola are very popular to be formulated in cosmetics or skin care products. Asiaticoside and madecassoside are well-known compounds contained in gotu kola that have an important role and are widely used in beauty products. However, the raw materials for these products are generally limited and relatively expensive. In addition, the established separation method used to isolate these compound are considered inefficient and environmentally unfriendly. This study was aimed to find the alternative methods used for separation asiaticoside and madecassoside that could be scaled up to industrial scale. Separation of asiaticoside and madecassoside was carried out using preparative HPLC and macroporous resin. Identification of asiaticoside and madecassoside using UPLC and Triplequadropole Mass Spectroscopy (MS/MS). The percentage of asiaticoside and madecassoside compound from saponin fraction using preparative HPLC were 41,95% and 28,38%, respectively. Meanwhile, by using macroporous resins, it was found that adsorption capacity for asiaticoside and madecassoside compounds by 8.1 mg/g dry resin (PAD 900) and 5.25 mg/g dry resin (PAD 610). Separation and purification of asiaticoside and madecassoside was successfully carried out by preparative HPLC. For separation using microporous resin, the static adsorption and desorption test showed that PAD 900 is better result compared to PAD 610. text
