Summary:Abstract: <br /> <br /> <br /> <br /> Pumpkin (Cucurbita pepo L., Cucurbitaceae) seed is usually consumed as food but previous researches showed that the seed is one of lignan sources which has various pharmacological activities, such as estrogenic activity. It was also reported that the study of lignan on male mice and human breast carcinoma cell line showed that lignan has anticancer activity against prostate and breast cancer, respectively. The purpose of this research, therefore, is to isolate lignan from pumpkin seed. Shelled pumpkin seed was manually separated from its hull and subjected to phytochemical screening and gas chromatographymass spectroscopy (GCMS). The ground crude drug was extracted by reflux using ethanol and the extract was fractionated by liquidliquid extraction using water and methylenchloride. The methylenchloride phase was fractionated by column chromatography and the lignan containing fractions were subjected to preparative thin layer chromatography (TLC). The isolate was examined for its purity by twodimensional TLC and was characterized by GCMS. The phytochemical screening showed the presence of steroid/triterpenoid and screening by GCMS showed the fragment of mass spectrum, m/z, 135, 151, 165, and 181, which are specific for lignan. One lignan has been isolated and tested for its purity by twodimensional TLC. <br />
