One major risk factor of atherosclerosis is hypercholesterolemia. Eleutherine americana (Aubl.) Merr, known as “bawang kapal” in Indonesia, has been used as folk medicine for cardioprotective activity. In this research, antihypercholesterolemic effect and acute toxicity test was investigated in Eleutherine americana (Aubl.) Merr. Model of hypercholesterolemic animals was achieved by giving a cockctail containing rich-cholesterol substances and propiltiourasilcontaining drinks for two weeks period. Induction was stopped with the beginning of drug therapy. Lipoprotein profile’s was observed in day 3, 7, and 14 of therapy. Acute toxicity test was observed on animal behaviour, lethality, and body weight of animals. Total cholesterol in all groups except controls were significantly decreased (p<0.05) since day-3 until day-14 of therapy. Water extract decreased total cholesterol from 105.21±38.86 mg/dL to 51.1±11.19 mg/dL. Ethanol-water extract decreased total cholesterol from 87.18±3.09 mg/dL to 44.8±8.1 mg/dL. Test substances did not influenced HDL. Triglyceride profile’s in all groups were fluctuative and did not show any significantly decrease. LDL significantly decreased (p<0.05) compared to positive control in all test groups. Therefore, Eleutherine americana (Aubl.) Merr could reduce total cholesterol and LDL level, meanwhile triglycerides and HDL were not affected. LD50 value of Eleutherine americana (Aubl.) Merr >3.6 g/kg BB. text
