SCREEN PRINTED CARBON ELECTRODE (SPCE) MODIFIED GRAPHENE/AUNPS CONJUGATED AMYLOID SERUM ANTIBODY A (SAA) AS A BIOMARKER OF CURE RATE OF COVID-19 PATIENTS

Covid-19 is a disease caused by a new virus, namely a new type of coronavirus (SARS-CoV-2). The disease can progress rapidly, leading to multiple organ failure and death. Therefore, it is necessary to develop effective, efficient, sensitive, and simple detection methods for monitor the cure rate. Bi...

The disease can progress rapidly, leading to multiple organ failure and death. Therefore, it is necessary to develop effective, efficient, sensitive, and simple detection methods for monitor the cure rate. Biomarker-based detection can be one of the alternatives in the test method. Amyloid A Serum protein is a potential biomarker in Covid-19 disease to monitor its infection rate. Amyloid A serum can be measured using voltammetry techniques. Determination of Amyloid Serum A was carried out using a modified Screen Printed Carbon Electrode (SPCE) biosensor with graphene/AuNPs conjugated against Amyloid A Serum Antibodies using the sandwich method. The morphology of the working electrode is characterized by Scanning Electron Microscopy-Energy Dispersive X-Ray (SEM-EDX). Determination of Amyloid Serum A on SPCE graphene/AuNPs conjugated antibodies Amyloid Serum A was measured in a solution of Posphate Buffer Saline contained Potassium ferrocyanide 10 ?M and 1 mM KCl. Optimization of antibody concentrations, measurement techniques, evaluation of peak currents and study of scan rates were carried out. The optimization results showed the best measurements using DPV and anodic peak currents decreased with SPCE graphene / AuNPs measurements compared to SPCE without modification (bare SPCE). The calibration curve shows a linear range at a concentration of 0.2 pM to 50 pM with a detection limit of 4.30 pM. In addition, SPCE graphene/AuNPs conjugated with Amyloid A Serum antibodies have good repoduction and good accuracy with a regain value of 99.93%. Spce graphene/AuNPs biosensors conjugated Amyloid A serum antibodies have good selectivity in the presence of common disruptive compounds present in blood samples such ascorbic acid, uric acid and glucose. Then the sample of Amyloid Serum A was measured using a nanodrop Spectrophotometer and the results showed no significant difference with voltammetry technique. So that the developed biosensor can be applied to the determination of Amyloid Serum A. text
