As a wild plant, the active compound content varies greatly depending on where it grows. Taking materials directly from nature without cultivation can cause a shortage of original plant sources so raw materials with quality that meet market demand are difficult to find. To overcome this problem, we need a method of cultivating plants with the availability of seeds with higher levels of nutritious substances. One way to get this plant is use plant tissue culture techniques. To improve the quality of tempuyung plants with plant tissue culture, an initial study is needed, namely the analysis of secondary metabolites content from the results of tempuyung tissue culture. This study aimed to determine the optimum concentration variation of growth regulators for growing callus and shoot cultures, along with analyze the content of secondary metabolites in tempuyung callus and shoot cultures. Leaves as explants were aseptically inoculated into Murashige and Skoog culture media and then incubated. Culture results were dried and then extracted by maceration with methanol. The extract was analyzed for its secondary metabolite content using TLC and UPLC-MS/MS. In callus induction, optimal callus formation was observed on MS4 (BAP 1 mg/L; 2,4-D 0,5 mg/L); in shoot induction, optimal shoot formation was observed on MT4 (BAP 1mg/L; NAA 0,5 mg/L). Based on the analysis using UPLC-MS/MS, in MS2 callus extract (BAP 0,5 mg/L; 2,4-D 1 mg/L) and MS4 rutin, luteolin, quercetin, and apigenin were detected. In MS3 shoots (BAP 1 mg/L; 2,4-D 1 mg/L) and crude extract of tempuyung leaves, rutin, hyperoside, luteolin, quercetin and apigenin were detected. In MT1 shoots (BAP 0,5 mg/L), MT2 (BAP 0,5 mg/L; NAA 0,5 mg/L), MT4, and MT5 (BAP 0,5 mg/L; NAA 1 mg/L) rutin, luteolin, quercetin, and apigenin were detected. In MT3t shoots (NAA 0,5 mg/L) hyperoside, luteolin, and apigenin were detected. In MT3k callus (NAA 0,5 mg/L) rutin, hyperoside and luteolin was observed. From the result of the analysis, it was observed that there were differences of metabolites content from callus and shoots cultured qualitatively. text |
