One of the arabica coffee producer areas in West Java is Sukabumi. However, chemical information on Sukabumi arabica coffee is still very limited in the literature. In this study, the metabolite profiles of green beans of arabica coffee obtained from various plantations in Sukabumi, including Ciayunan, Pondok Halimun, and Selabintana were evaluated by 1H NMR-based metabolomics. Samples of ground green coffee beans were extracted with D2O and measured by NMR spectroscopy. The obtained 1H NMR spectra were processed and evaluated with multivariate data analysis using Orthogonal Partial Least Square Discriminant Analysis (OPLS-DA) as the main model. 19 metabolites were successfully identified on the 1H NMR spectra, including caffeine, trigonelline, sucrose, chlorogenic acid, quinic acid, malic acid, lactic acid, and fatty acids. The concentrations of several identified metabolites were determined semi-quantitatively using 1H NMR analysis techniques. For example, Pondok Halimun coffee had a higher concentration of caffeine than other coffee samples with a concentration of 31.59 mM. The score plot of the OPLS-DA model was successful in classifying the metabolite profiles of the coffee samples based on their geographical origin. Loading plot analysis showed the signals belonging to fatty acids, sucrose, trigonelline, chlorogenic acid, lactic acid, and quinic acid, contributing to the classification of coffee samples based on their geographical origin. In this study, 1H NMR-based metabolomics has succeeded in revealing the differences in the metabolite profiles of Sukabumi Arabica coffee beans cultivated in different areas. text |
