The aim of the present study was to formulate tamanu oil into an optimal and stable Nanostructured Lipid Carriers (NLC) to enhance its activity against the acne-causing bacteria Cutibacterium acnes. The formulation of NLC-Tamanu Oil was optimized using a Box-Behnken Design with solid lipid to liquid lipid ratio (X1), surfactant (X2), and cosurfactant (X3) as independent variables, and particle size (Y1) and polydispersity index (Y2) as response variables. In vitro antibacterial activity testing was performed using the broth microdilution method. The NLC-Tamanu Oil formula was successfully prepared with a particle size of 281.13 ± 7.32 nm, polydispersity index of 0.301 ± 0.055, zeta potential -19.44 mV, entrapment efficiency of 99.54 ± 0.38%, and pH of 4.99 ± 0.12. In vitro testing results showed that encapsulating tamanu oil into NLC increased its antibacterial activity by more than 2-fold, from an IC50 value of 1.74 µg/mL to 0.76 µg/mL, classifying it as a strong antibacterial agent. NLC-Tamanu Oil was most stable when stored at 4°C after one month of testing. Based on the findings of this research, it can be concluded that formulating NLC could enhance the antibacterial activity of tamanu oil against the acne-causing bacteria C. acnes. text |
