Black bangle (Zingiber ottensii Val.) is a plant that has the potential to inhibit ????-glucosidase enzyme. This study aimed determine the inhibitory potential of bangle hitam (Zingiber ottensii ??????íð???± ±??? ®??????»? ®?ð??????? ?¸ ð??? ?±? ã ð???????- glucosidase enzymes. Using acarbose as a positive control?????±??ð?ã ?í± ð?íðð???? ®?ð?ð???» ?í± ????- glucosidase enzyme was examined in vitro using the colorimetric method at a wavelength of 405 nm. Bangle hitam (Zingiber ottensii Val.) rhizome simplicia was extracted by the maceration method using 96% ethanol solvent, then extracted liquid-liquid using n-hexane and ethyl acetate solvents. The IC50 values of the ethanol extract, n-hexane fraction, ethyl acetate fraction, and water fraction, were 99.64 ± 3.52 µg/mL, 72.98 ± 1.65 µg/mL, 108.13 ± 2,89 µg/mL, and 696.59 µg/mL respectively. Advanced fractionation was performed on the n-hexane fraction using the classical column chromatography method and followed by purification using preparative thin-layer chromatography. The IC50 value of the isolate and acarbose result were 323.53 ± 22.74 µg/mL and 45.41 ± 0.52 µg/mL. Based on the characterization using a specific spot viewer and two-dimensional paper chromatography, it is known that the isolate is a compound of the flavonoid group belonging to the isoflavone subgroup. text |
