Immune response can be optimize using immunomodulators. Immunomodulators are compounds
that can regulate the immune system. Immunomodulators can be obtained from plants as sought
in this study. This study aims to examine the immunomodulatory effect of ethanolic extract of green
tea leaves. The active compounds in green tea leaves were extracted by maceration using 70%
ethanol. Two doses of green tea leaves ethanolic extract (EDTH) tested were 0.3 mg/kg bw (EDTH
DR) and 0.6 mg/kg bw (EDTH DT). The immunomodulatory activity of EDTH was tested on BALB/c
mice. Test results on non-specific immune response through carbon clearance test showed that
EDTH DR and DT were immunostimulant with phagocytic index EDTH DR = 1.65 and EDTH DT = 1.14.
Both doses of EDTH significantly increase the liver index (DR = 6.2 ± 0.9 and DT 6.3 ± 0.3 vs. control
5.4 ± 0.4). At low doses EDTH increase the spleen index significantly (1.02 ± 0.3 vs. 0.64 ± 0.04). In
the specific immune response test, both doses of EDTH stimulated the humoral immune response by
increasing primary and secondary antibody titre. Both doses of EDTH also affected the cellular
immune response by increasing the production of IFN-?, and only EDTH DT significantly increased IL2 production (8,806 ± 0,597 vs 6,101 ± 0,793 pg/mL). In the lipid peroxidation test, both doses of
EDTH could inhibit the formation of MDA as an indication of the antioxidant effect. Based on the
results of this study it can be concluded that ethanolic extract of green tea leaves has an
immunomodulatory effect with immunostimulant activity.
