Acne therapy using antibiotics often causes bacterial resistance. Therefore, the search for alternative treatments for acne derived from natural plant materials is an interesting matter. This study aims to evaluate the antibacterial activity of extracts and fractions from Tamanu seeds (Calophyllum inophyllum L.) against acne-causing bacteria, Propionibacterium acnes ATCC 11827, and Staphylococcus epidermidis ATCC 12228. Tamanu seeds were extracted using maceration method with ethanol 70% as solvent. Fractionation of the extract was performed using vacuum liquid chromatography (VLC) with eluent composition of n-hexane, ethyl acetate, and methanol as solvents. The screening method used for activity testing was agar diffusion method by determining the diameter of inhibition zones, and the minimum inhibitory concentration (MIC) was determined using microdilution method. The results showed that the MIC of the extract, fractions 3, 4, 5, and 6 from Tamanu seeds against Propionibacterium acnes ATCC 11827 were 0.5; 1.0; 1.0; 31.25 and 62.50 µg/mL, respectively. The MIC of the extract, fractions 3, 4, 5, and 6 from Tamanu seeds against Staphylococcus epidermidis ATCC 12228 were 62.50; 15.63; 15.63; 31.25 and 250 µg/mL, respectively. Therefore, tamanu seed extracts and fractions has potential activity against acnecausing bacteria. text
