Water spinach leaves are believed to induce drowsiness, while chamomile tea is commonly consumed to improve sleep quality. Therefore, this study was conducted to test the hypnotic and sedative effects of a combination of water spinach leaf infusion and chamomile flower. The testing was performed on male Swiss-Webster mice divided into five groups: a control group receiving distilled water, a reference group receiving diazepam (3 mg/kg body weight), and three test groups (I, II, and III) receiving water spinach leaf infusion (1,3 grams/kg body weight), chamomile leaf infusion (1,3 grams/kg body weight), and a combination of water spinach leaf infusion (650 mg/kg body weight) with chamomile flower (650 mg/kg body weight), respectively. The sedative effects were tested through behavioral experiments and sedation rating scale measurements. The hypnotic effects were evaluated by measuring four parameters: sleep latency, sleep duration, sleep efficiency, and brain and dorsal temperature measurements in mice induced to sleep with ketamine (200 mg/kg body weight). The results of the sedative effects test showed that the combination group provided a stronger sedative effect than the individual groups in all four parameters, including reduced motor activity, ptosis effect, decreased alertness, and sedation rating scale average. The results of the hypnotic effects test indicated that the combination group (95,71 ± 1,34%) exhibited a higher mean sleep efficiency compared to both individual infusions and showed no significant difference compared to the diazepam group (97,48 ± 0,9%) and the single water spinach group (94,81 ± 1,39%). In conclusion, the combination of water spinach leaf infusion and chamomile flower can provide better sedative and hypnotic effects compared to the individual infusions. text |
