Of the many benefits, hesperidin has the potential to be utilized optimally as a drug and supplement. Meanwhile research and production of this compound is still limited by the methods commonly used with small recoveries and it does not have good repeatability. So efforts are needed to get these compounds in sufficient quantities. This research was conducted to develop a purification method for the hesperidin flavonoid compound using the extraction method of boiling with water and fractionation using polymethacrylate resin. Method development is carried out by determining the parameters of linearity, specificity, limits of detection and quantification, as well as accuracy and precision. The results obtained from the development of the method are linearity showing R 2 = 0.9993, specificity indicating the absence of impurities at the time the retention of the target compound appears, precision accuracy that is within the acceptance range, as well as the limit of detection (LOD) and quantification (LOQ) were 0.14 and 0.42 ppm respectively. The extract was then fractionated using macroporous resin and the results obtained were then analyzed by HPLC (High Pressure Liquid Chromatography). The resin elution fraction showed an increase in hesperidin levels from 11% to 28% in elution using 70% ethanol and 18% in 96% ethanol elution. text |
