ISOLATION, CHARACTERIZATION AND IDENTIFICATION OF THE THERMOPHILIC BACTERIA PRODUCING LIPASE

Abstract: <br /> <br /> <br /> <br /> <br /> Lipases (triacylglycerol acylhydrolases, EC 3.1.1.3) catalyze the hydrolysis of water-insoluble esters and triglycerides at the interface between the insoluble substrate and water. Thermophilic bacteria producing lipase were isolated from Pasir Impun thermogenic composts at temperatures between 50 and 7 farenheit C. From 40 isolates tested only 10 isolates showed lipolytic activity on agar plate media containing olive oil and rhodamine B. The isolates showed lipase activity in the range of 15 to 315 U/L on p-nitrophenyl palmitate (pNPP) as substrate. Two isolates, namely 6A and 13C, were further characterized and showed maximum specific lipolytic activity at a mid exponential phase. Chromosomal DNA from these isolates were isolated and the 16S rRNA genes were successfully amplified resulting a fragment with the base length was around 1.5 kb. Analysis of the 16S rRNA genes sequence using homological and phylogenetical methods concluded that the Pasir Impun 6A and 13C isolates were close to Bacillus licheniformis and Bacillus subtilis, respectively.
