Summary:Stevia (Stevia rebaudiana Bertoni) is a plant producing the sweet compound steviol glycosides with the main compound stevioside, whose leaves have been used as a natural low-calorie sweetener and have various health benefits and industrial applications, thus attracting the attention of consumers. Research studying stevioside purification in aromatic macroporous resin columns is still limited. This research was conducted to purify stevioside compounds using aromatic macroporous resins through dynamic adsorption and desorption methods in a resin column. The results show that with a flow rate of 3 BV/hour, 573.3 mg of stevioside could be adsorbed on 25 g of wet aromatic macroporous resin. Stevioside and impurity compounds could be desorbed with gradient elution using 50-70% and 0-70% of ethanol respectively. The purity of the stevioside compound increased from 22.65% (w/w) to 62.68% (w/w) after being eluted in a gradient using 0-70% ethanol, and 22.65 ± 0.44% to 27.93 ± 0.87% (w/w) after being eluted using 4 BV of 50% ethanol. The purity of stevioside compounds increased after being adsorbed and desorbed in an aromatic macroporous resin column. A more optimal elution system and other purification efforts are needed to obtain a higher purity of stevioside.
