Ectoine is one of the bioactive compounds in cosmeceutical products due to its skin health benefits. Ectoine can be produced by Virgibacillus salarius as response to high salinity. This research aims to determine the growth kinetics and ectoine production of Virgibacillus salarius at NaCl concentrations 2% and 10% (w/v). Virgibacillus salarius cultivation is done for 48 hours in Zobell medium with cultivation conditions such as inoculum concentration of 105 CFU/mL, 150 rpm agitation, and 30? temperature. Ectoine is obtained from the cells by bacterial milking with 1,5% NaCl (w/v). NaCl concentration of 10% (w/v) resulted in the best growth for Virgibacillus salarius in which the exponential phase is at 3-39 hours. Concentration of substrate declines with growth phase while ectoine was produced at the end of exponential phase and stationary phase with the highest concentration as much as 6,82 mg/L. Specific growth rate and generation time of Virgibacillus salarius at 10% (w/v) salinity consecutively are 0,70±0,01/hour dan 1,00±0,02 hour. The YP/X values of Virgibacillus salarius at 10% (w/v) salinity is 4,44x10-3 ±1,45x10-3 g ectoine/g cell dry weight. The increase in medium salinity from 2% to 10% (w/v) can also increase Virgibacillus salarius ectoine production 153,27-fold. Ectoine productivity from Virgibacillus salarius at 10% (w/v) salinity consecutively is 1,23x10-4±2,75x10-5 g/L/hours. text
