Summary: | <p align="justify"> Amino acids are molecules that make up proteins through peptide bonds. Protein has many important roles in living things. Black soldier fly larvae (BSFL) are a source of biomass which has a high protein content. Glycine is one of the amino acids that has many benefits and can be increased through solid-state fermentation of BSFL with Lactobacillus acidophilus. This study aims to determine the effect of fermentation on crude protein content in larvae, protein recovery, Degree of Hydrolysis (DH) in larval protein hydrolysate, glycine content, and amino acid composition in larval fractionated protein. In addition, it also determines the effect of chromatographic flow rate on fractionated protein concentration. The crude protein content in the fermented sample was 34.67%, which was 2.4% higher than the control sample. Protein recovery of protein extract with alkaline extraction using 0.25 M NaOH was higher on the control sample with a value of 41.68%. DH of hydrolyzed protein extract with 3% bromelain enzyme was also higher on the control sample at 53.19%. Protein hydrolysate fractionation using Size-Exclusion Chromatography with three flow rates (0.5;1;1.5 mL/min) resulted in protein concentration between 1,080-1,394 mg/mL on the control sample and 1,334-1,366 mg/mL on the fermented sample. The composition of the amino acids in the fermented protein hydrolysatewas different from the control sample such as serine and aspartic acid content. The glycine content obtained using Ultra High-Performance Liquid Chromatography Photodiode Array in the fermented sample was 4.3% higher, which was 0.325 mg/g fraction compared to the control sample which was 0.312 mg/g fraction.
