However, conventional stevia cultivation on a big scale is considered difficult, so the proposed solution is the application of tissue culture method in TIS RITA® bioreactor. The purpose of this study is to determine how applying methyl jasmonate (MeJA) elicitor at a concentration of 100 ?M affects the growth and production of stevioside and rebaudioside-A in stevia shoot culture. When added to the culture media, MeJA has been shown to improve the the production of secondary metabolites in plants, notably steviol glycosides in stevia. MeJA had a positive impact on the synthesis of stevioside and rebaudioside-A in stevia shoot culture, with values of 59.93 mg/g dw and 0.0077 mg/g dw, respectively, as compared to the control, which had values of 58.70 mg/g dw and 0.0075 mg/g dw. The parameters of Relative Growth Rate (RGR), height increment, and number of shoots, which were 0,096 g/day, 0.633 cm, and 2.93-fold, respectively, showed no significant effect on the growth of stevia shoot culture. The culture media's pH, sugar content, and conductivity value all decreased, demonstrating the presence of nutrient biotransformation activity by the stevia shoot culture. As a result, the findings of this study suggest that the TIS RITA® bioreactor and MeJA could be used to produce stevia plants with high steviol glycoside content in large quantities rapidly in the future. text
