Summary:Diabetes mellitus (DM) is a chronic disease which is the highest contributor to death in Indonesia due to excessive consumption of sugar. Steviol glycosides (GS) in Stevia rebaudiana Bertoni can be used as a zero-calorie alternative natural sweetener because it has a sweetness 250-400 higher than sucrose or granulated sugar. Cultivation of Stevia rebaudiana Bertoni is naturally considered inefficient, therefore an alternative method of in vitro propagation is needed in a mist bioreactor in order to increase the rate of multiplication and growth, also accumulation of GS in stevia culture. Elicitation using high concentrations of methyl jasmonate (MeJA) in a short time can also increase the rate of GS biosynthesis. The aim of this study was to determine the effect of 100 ?M MeJA elicitation on plant growth and the content of stevioside and rebaudioside-A in the Stevia rebaudiana Bertoni culture which was cultivated in a mist bioreactor for 14 days. Elicitation for 24 hours increased culture height 1,17 times, shoot number 1,18 times, total biomass 1,1 times, and relative growth rate 1,25 times higher than control culture. Stevioside and rebaudioside-A obtained in dry weight (DW) GS increased in cultures treated with MeJA. Stevioside from control culture and MeJA 100 ?M treatment were 62.82 mg/g DW and 68.53 mg/g DW respectively, while rebaudioside-A was 0.0104 mg/g DW and 0.0113 mg/g DW. So, it can be concluded that the elicitation of MeJA 100 ?M for 24 hours increased the growth and content of stevioside and rebaudioside-A in the Stevia rebaudiana Bertoni culture that cultivated in the mist bioreactor.
