Production of B lactam antibiotics is known to be influenced by the presence of carbohydrate, nitrogen and phosphate. In many antibiotics fermentation system it has been reported that the presence of phosphate in the culture medium strongly affected both cell growth and synthesis of products. In this research the application of corn steep liquor which contain phosphate compounds and the quantity of celite as supporting immobilized spore of Penicillium chrysogenum ATCC 26818 has been evaluated. The concentration of corn steep liquor were varied: 1.5; 2.0; 2.5; 3.0 and 5.0% and the quantity of celite were varied : 2.5; 5.0; 7.5 and 10.0g for 25.0 mL spore suspension that contain 10e8-10e9 spora/mL The concentration of penicillin in the fermented broth was determined by both biochemical assay and HPLC method. S. aureus was used as test micro-organism and acetonitril with KH2-PO4 (68:32) as an eluen. The optimum product was obtained in using 2.5% of corn steep liquor and 7.5g of celite. <br />
