Summary:Tin (Ficus carica L. L.) is a high nutritional value fruit producing plant that is often used to treat various diseases. The demand for tin fruit products in the world is very high so the development of tin fruit production in Indonesia is very promising. To be able to develop tin fruit production, an efficient and effective seedling production optimisation system is needed. Tissue culture technique using Ebb-and-Flow bioreactor is suitable for optimising seedling production because it can increase the transfer of nutrients and gas to the explants. This study aims to optimise soaking time on seedling production time and determine the best soaking time in tin explant propagation by analysing the multiplication rate, relative growth rate, height growth rate, nutrient consumption, and survavibility rate of tin explants. Tin explants were cultivated on half-strength liquid Murashige & Skoog medium, 1.13 ppm BAP, and 0.5 ppm GA for 14 days using an Ebb-and-Flow bioreactor with variations in soaking times of 20 and 30 minutes every 6 hours and using a continuous soaking system. Based on the research, the results of the analysis of tin explants cultivated in the Ebb-and-Flow bioreactor with immersion times of 20 and 30 minutes every 6 hours had multiplication rates of 14.76 and 12.58 shoots/explanant, relative growth rates of 0.09 and 0.06 g/g.day, height growth rates of 1.02 and 0.72 mm/day, nutrient consumption rates of 9.05% and 10.55%, and survivability rates of 92.31 and 87.50%, respectively. Meanwhile, tin explants cultivated in the continuous immersion system had a multiplication rate of 4.50 shoots/explanant, relative growth rate of 0.04 g/g.day, high growth rate of 0.56 mm/day, nutrient consumption of 15.08%, and survavibility rate of 72.73%. From the results of this research and analysis, the cultivation system using Ebb-and-Flow bioreactor with soaking time of 20 minutes every 6 hours is the best system in tin seedling production.
