Wound is an example of skin damaged. Based on previous studies, lavender and rice milk extract
showed the wound healing potential. This study aimed to investigated the potential cytotoxic effect of
the extracts on the NIH 3T3 cell line and in vitro skin scratch assay also carried out to study the
mechanism of wound healing from the lavender and rice milk extract. The cytotoxicity assay was carried
out using Presto Blue reagent. Lavender extract tested was prepared in School Pharmacy of Intitut
Teknologi Bandung (SF extract) and extract product on the market (KM extract) with 0,5% and 1%
concentration for the assay. Rice milk extract tested was prepared in School Pharmacy of Intitut
Teknologi Bandung (SF extract) and extract product on the market (KM extract) with 0,5% and 0,75%
concentration for the assay. The results of the cytotoxicity assay using Presto Blue show lavender and
rice milk extract are not cytotoxic to cell because the decreased of cell viability was less than 30% (ISO,
2009). However, this result was not relevant with microscopy observations from morphology of that
incubated with rice milk extract. Therefore, an additional cytotoxicity assay was carried out with Trypan
Blue and it was found that rice milk sample was cytotoxic because the decreased of cell viability was
more than 30%. Skin scratch assay results showed the wound closure percentage of the cells treated
with the lavender and rice milk extract was below 10% and cell death occurred was observed after
incubation with several extract concentrations, especially using the rice milk extracts. Based on these
result, the in vitro skin scratch assay method was not suitable to study the wound healing activity of
lavender and rice milk extract.
