Summary:Porang (Amorphophallus muelleri Blume) is a wild plant which is valued as a commodity that has high economic value. Porang plants contain a glucomannan compound which accumulates a lot in the tubers. Glucomannan has been widely used in industry and pharmaceuticals because of its distinctive characteristics of being soluble in water and being able to swell quite large by forming a thick mass. The aim of this study was to isolate the glucomannan compounds in porang tubers with various solvents and determine the levels of each glucomannan extract, as well as conduct as conduct a preliminary study on the preparation of a combination of glucomannan powder and grass jelly leaf tablets. Isolation of glucomannan was carried out using two methods, namely the purification method using 96% ethanol and the extraction method using two different solvents, namely water and aluminum sulfate. Prior to the extraction process, a preliminary test was carried out to determine the particle size of porang tubers which contain a lot of glucomannan compounds by means of multilevel sieving, then continued to determine the swelling index. Porang tuber powder particle size of 60 mesh was chosen because it has the highest swelling index which is at 8.33 ± 0.47 mL/gram. The content of glucomannan compounds purified with 96% ethanol and extracted with water and aluminum sulfate 0,3% was determined using the DNS (dinitrosalicylic acid) colorimetric method with a UV-Vis spectrophotometer at 550 nm. The result of glucomannan compounds level in porang tuber simplicia was 53.93 ± 3.73%, 96% ethanol extract glucomannan was 48.34 ± 2.10%, glucomannan aqueous extract was 64.82 ± 2.09%, and glucomannan aluminum sulfate extract was 82.97 ± 1.02%. There is an anomaly in the data on the content of the 96% ethanol extract of the glucomannan compound because it is smaller than the porang tuber simplicia before being given treatment. In the manufacture of glucomannan powder tablets and grass jelly leaves powder tablets, must take a reformulate, so it become tablets that have better quality.
