Summary:The use of groundwater as a source of clean water in rural communities in Indonesia is still quite high with water quality conditions that do not meet the quality standards as clean water or drinking water. This study was conducted in Kalierang Village, Bumiayu District, Brebes Regency, Central Java wicth the aim of evaluating water quality microbiologically by measuring E. coli and Total coliform in non-PDAM clean water sources. The measurements of E. coli and total coliform in 40 grundwater samples in Kalierang Village were conducted using the membrane filter method, and questionnaires were distributed to determine the use of water sources, and sanitation. Data analysis used descriptive analysis, Spearman correlation, and QMRA. The test results of groundwater such as shallow well, and deep well in Kalierang Village were generally positive for Total coliform and E. coli exceeded the quality standards of Permenkes No. 2 of 2023. There is a significant correlation between variable well depth (p-value = 0,000), distance of sanitation (p-value = 0.000 and 0,003) with contamination of E. coli and Total coliform. The result of QMRA (Quantitative Microbial Risk Assessment) showed a possible risk of E. coli infection during wudhu (1,4x10-5 ± 1,2x10-5 ), and tooth brushing (5,5x10-6 ± 4,8x10-6 ). As for bathing (2x10-7 ± 0,00), it shows a low risk of E. coli infection.
