Curcumin is estimated to potentially reduce alcohol dependent because of its role in arachidonic acid pathway. This study aimed to determine the effect of curcumin in animal models of alcohol dependent. Experiment were conducted using Conditioned Place Preference (CPP) paradigm, which consists of four stages, habituation, conditioning, abstinence, and relapse. Habituation conducted on space experiments, treatment and CPP apparatus. In the conditioning phase, the test animals were given curcumin and alcohol. At abstinence phase, the test animals were given only saline and at the relapse stage given a priming dose of alcohol. Acetylcholinesterase (AChE) activity measurements were performed at the end of each behavioral tests using Ellman's method. Administration of curcumin dose of 1 mg/kg and 3.2 mg/kg showed a significant difference of preference score compared to the saline group and the comparison group (p <0.05), while the curcumin dose of 10 mg/kg did not show significant differences to the saline group and comparison group. AChE activities of curcumin dose group (1; 3,2; 10) mg/kg at post-conditioning test were significantly different compared to the positive control group (p <0.01). However only curcumin dose of 10 mg/kg were not significantly different compared to the saline group. While the relapse phase, a significant difference (p <0.01) compared to the positive control group was only demonstrated by curcumin 10 mg/kg, saline, and comparison group. Based on the experiment results, administration of curcumin at dose 10 mg/kg during conditioning phase and before the priming dose of alcohol can lower preference scores on post-conditioning and relapse test in line with the decreasing in AChE activities.
