Its treatment becomes complicated and the cost is also expensive. Pepper elder has bioactive component which acts like estrogen and is proved able to improve microstructure of bone. The objective of research was to investigate the effect of pepper elder herbs juice and ethanol extract on OVXinduced osteoporostic rat. Osteoporosis was inducted by ovariectomy with double dorsolateral approach on female Wistar rat. The animals were undergone SHAM (normal group) and ovariectomy surgery. The ovariectomized rats divided into 6 groups treated with CMC Na 0,3% (control group), ethynilestradiol 4.5yg/kg bw, juice 50 mg/kg bw, juice 100 mg/kg bw, ethanol extract 50 mg/kg bw, and ethanol extract 100 mg/kg bw. The rats was treated for 6 weeks started from one week after operation. Weight gain, uterus index, serum and urine calcium level, and serum ALP level were measured. Three dimensional image and quantitative parameters of femur from scanning using microCT scanner were conducted. All of pepper elder-treated groups did not show activity in weight gain reduction and uterus index significantly to control group (p<0.05). Rats treated with ethanol extract at 100 mg/kg bw significantly decreased QLP serum level and reduced excretion of urine calcium compared to control group (p<O,05). This changes did not showed significantly different to normal group (p<O.05). Furthermore, ethanol extract 100 mg/kg bw-treated group showed improvement on three-dimensional image of trabecular bone compared with control group. That group also counteracted the reduction of bone volume fraction, trabecular thickness and number, and connectivity index. Ethanol extract of pepper elder herbs at dose 100 mg/kg bw had preventive effect on OVX-induced osteoporotic rats. text |
