There are some factors which is still a challenge in glucosamine as a drug development : large dose to absorbed in a transdermal route and maillard product easily produced when glucosamine soluble in water. This research aims to develop glucosamine HCI for skin application by enhancing the permeability toward stratum corneum and the stability of drug. In this research glucosamine HCI developed into microemulsion where glucosamine soluble iff water would be on the continuous phase while the oil which has small lobul size as a dispersed phase. This microemulsion also act in preventing the interaction among the free molecules water initiating oxidation. The optimum value of microemulsion formula were glucosamine hydrochloride 15%, isopropyl myristate 4,2%, Tween 80 22, 7%, isopropyl alcohol 5, 7%, sodium metabisulfite 0, 1% and deionized water 52,3%. Microemulsion then formulated into cream preparation by using polyethylene glycol 1000, cetostearyl alcohol, liquid paraffin and cera alba. This preparation then tested for stability at temperature 25 0C and 400C for 28 days. Residual levels analysis using High Performance Liquid Chromatography (HPLC) showed its stability at 102,69% 0,58 for temperature 25 0C; 102,480/0 0,77 for temperature 400 C (P>0,05). The pH decreased, the viscosity increased for both temperature. In vi[ro diffusion study using Franz-type diffusion cells with a snake skin membrane (Phyton reticularis) as a model of permeation, showed that microemulsion cream of glucosamine HCI give the highest amount of diffusion (857,54 ± 0,06 gg/cm ) rather than glucosamine in cream A (722,99 ± (),80 gg/cm ) and simulation O/W cream glucosamine HCI 10% (152,95 0,25 ug/cm ). In vivo irritation study were using three male New Zealand albino rabbits. The results showed that no erythema and no oedema after application of microemulsion cream (primary irritation index ¯ 0). text
