Summary: | Gout is a common of rheumatic disease that usually happened by increasing of uric acid (hyperuricemia). Previous study revealed that sidaguri leaves and red ginger leaves had potency as antihyperuricemia. The aims of this research were to study antihyperuricemic activity of sidaguri leaves,red ginger leaves, and their combination using xanthine oxidase assay, the ability to decrease uric acid level in serum, and uricosuric effect in hyperuricemic rats. Sidaguri leaves and red ginger leaves were extracted by reflux using 96% ethanol and evaporated by rotavapor. Chromatogram pattern of extract was perfomed by thin layer chromatography (TLC). 'The inhibitory effect of xanthine oxidase activity was determined by UVVis spectrophotometry. Antihyperuricemic activity study used male Wistar rats, the animals were hyperuricemia induced by potassium oxonate (250 mg/kg bw ) and high purine diet. After administrating by extract, uric acid level in serum and urin (uricosuric activity) was compared to allopurinol and probenesid. 1050 of red ginger leaves, sidaguri leaves, and its combination were 19.083 ppm, 7.809 ppm, dan 1.71 ppm. IC50 of allopurinol as comparative was 0.189 ppm. Xanthine oxidase inhibitory activity assay of its combination had additive properties. Sidaguri extract at dose of 100 mg/kg bw and extract red ginger leaves at dose of 100 mg/kg bw were suspected had high uricosuric activity to decreasing uric acid level in serum, however sidaguri extract at dose of 50 mg/kg bw could decrease uric acid level in serum by xanthine oxidase inhibitory activity.
