Background : Cardiovascular disease is a main cause of death in the world. Research of herbal medicine to handling the disease indicate escalating. Therefore, an experiment of vasodilation effect of ethanolic extract of Anredera cordifolia (Ten) V. Steenis , ethanolic extract of Sonchus aryensis , ursolic acid, oleanolic acid and apigenin om isolated rabbit aortic and frog heart were done. The objective of the experiment was to determine Of vasodilation effect and the mechanism of action involved . Methods : Aortic rabbit were placed in organ bath and precontracted with norepinephrine (2.9 x 10-3) and potassium chloride (40gM) in discrete manner and then followed by the test compound (ethanolic extract of Anredera cordifolia (Ten)V. Steenis , ethanolic extract of Sonchus arvensis , ursolic acid, oleanolic acid and apigenin) . An anesthetized frog that exposes of the heart was precontracted with norepinephrine (2.9 x 10-3) . The vasodilation responses in isolated rabbit aortic were evaluated through time duration to reach 100% relaxation when the precontracted returned to the baseline position . Whereas the vasodilation response in frog heart was evaluated through reducing of heartbeat . Results : Ethanolic extract of Anredera cordifolia (Ten) V. Steenis (0.9 mg/ml) produced significant vasodilation effect (p<0.05) of the norepinephrine precontracted rabbit aortic rings . In addition , produced significant vasodilation effect (p<0.05) of the potassium chloride precontracted rabbit aortic rings . The vasodilation effect that produced by ethanolic extract of Anredera cordifölia (Ten) V. Steenis through inhibition of channel calcium and NO , since the pretreatment of isolated rabbit aortic rings with addition methylene blue inhibited the NO mediated vasodilation . In addition , ethanolic extract Afterward , the ethanolic extract of Anredera cordifolia (Ten) V. Steenis showed reduction of the heartbeat that mediated through beta-I receptor inhibition Whereas ethanolic extract of Sonchus arvensis (0.9 mg/ml) produced vasoconstriction of the norepinephrine and potassium chloride precontracted aortic rabbit rings . After this , the experiment continued for the secondary metabolite contents from ethanolic extract of Anredera cordifolia (Ten) V. Steenis. (ursolic acid, oleanolic acid and apigenin) that believed have benefit effect
Ursolic acid (0.5gg/ml) not produced vasodilation effect of norepinephrine and
potassium chloride precontracted aortic rabbit . But , this ursolic acid (0.5gg/ml) produced vasodilation effect that characterized by reduction of the heartbeat. This results showed ursolic acid had the role in reduction of the heartbeat that produced of ethanolic extract of Anredera cordifolia (Ten) V. Steenis Whereas for oleanolic acid (0.5 gg/ml) showed significant vasodilation effect (p<0.05) of the norepinephrine precontracted , but not showed vasodilation effect in potassium chloride and norepinephrine plus methylene blue precontracted . In addition , this oleanolic acid showed reduction of the heartbeat . This results indicated if vasodilation effect of oleanolic acid through NO and inhibition of beta-I . For the apigenin experiment , the apigenin (0.05 gg/ml) showed significant vasodilation (p<0.05) of norepinephrine , methylene blue and norepinephrine and potassium chloride precontracted . Moreover , the apigenin showed reduction of the heartbeat . This results indicated if vasodilation effect of apigenin through inhibition of calcium channel and inhibition of beta-I receptor. Conclusion : The experiment results is ethanolic extract of Sonchus arvensis produced vasocontriction of the aortic rabbit rings . Whereas ethanolic extract of Anredera cordifolia (Ten)V. Steenis, ursolic acid , oleanolic acid and apigenin produed vasodilation effect through NO effect, inhibition of calcium channel, inhibition of beta-I receptor, and inhibition beta-I receptor, and NO effect, inhibition beta-I receptor , and inhibition of calcium channel , inhibition of beta-I receptor respectively
