However, there is still very limited information regarding men contraceptives, it led to unsuccessful KB program. Gandarussa (Justicia gendarussa. Bunn.f.) is a plant in Indonesia, which has been used by the people of Papua as a contraceptive drug for men. This plant contains gandarusin A, a compound that has antifertility activity. It works through prevention of penetration of spermatozoa by lowering the activity of enzyme hyaluronidase. The time needed to get gandarusin A from wild type is quite long so the use of tissue culture to produce gandarusin A is very potential. The aim of this research was to analyze gandatusin A of J. gendarussa Burm.f in vitro culture. Methods : The axillary (shoot) pan of gandarussa was inducted to grow as buds in MS (Murashige dan Skoog) media, WPM (Woody Plant Medium) and B5 (Gamborg) with IBA and BAP hormone. Afterward, the optimation of hormone was done to MS and WPM media to obtain the optimal growth. The in vitro leaf of gandarusa was inducted to form callus using a suitable hormone and was optimated to obtain friable callus. The friable callus was subjected to MS and WPM media to obtain a cell suspension which counted for its growth index. The gandarusin A level in all in vitro culture was analyzed using spectrophotodensitometry using TLC scanner. Result : The induction of Gandarussa in vitro shoot was successfully conducted for 8 weeks. WPM media containing IBA+BAP (0, 1 + 1) mg/L is inducting the shoots better than both MS and B5. The optimization of hormones shows that the average growth in MS media with homone NAA+BAP (0.3 ± 0.03) mg Lis better than the IBA + BAP (0.3 ± 0.03) mg /L while the media WPM by the use of IBA + BAP (0.3 + 0.02) mg / L is better than the NAA + BAP (0.3 + 0.02) mg / L. There is a significant growth difference between media with and without additional vitamin. The WPM medium which contains more vitamins (5 x) has the best growth index (23.88 ± 0.26). Gandarusin A was detected at Rf 0.25 in in vitro culture of Gandarusa leaves and was not detected in callus culture.
