It has been widely known as a plantation crop, especially as food. Traditionally, the stem bark of this plant used as mouthwash. The major compounds of cashew stem bark are terpenoids and phenolic compounds. However, the phytochemical study of compounds was still limited. In this study, antibacterial compounds were isolated and their structures were determined. The isolation was done through series of separation methods include extraction, fractionation, purification, guided by antibacterial assay. Extraction was performed by maceration, followed by fractionation and purification by vacuum liquid chromatography and radial chromatography. Antibacterial activity of the extract and fractions was monitored by disc diffusion method and micro dilution techniques. The purity of the isolated compounds was monitored using two-dimensional thin layer chromatography. The chemical structure of the isolated compounds were elucidated using spectroscopic data, included UV-Vis, IR, 1H-NMR, 13C-NMR , and 2D-NMR (HSQC and HMBC). Two compounds were isolated from cashew nuts stem bark as yellowish crystals (1) and a yellow amorphous powder (2) that were identified as pinostrobin (1) and pinocembrin (2), with minimum inhibitory concentration values were 500 ppm and 125 ppm, respectively. text |
