Summary: | Nigella sativa Linn, (family Ranunculaceae) seeds is known to have a strong anti inflammation effect. The compound which has responsibility to perform this anti inflammation effect is thymoquinone, one of the compound contained in volatile constituents of Nigella sativa oil, but later it was found that even the fixed oil of Nigella sativa oil has a greater anti inflammation effect than thymoquinone. Limited usage of Nigella sativa in the form of oil or seeds powder have a problem in bioavailability, makes it less popular in therapeutic purposes. To overcome this problem, the self nanoemulsion dossage form is developed to improve the activity of Nigella sativa oil as anti antiinlammatory agent. Nanoemulsion of Nigella sativa oil is made by spontaneous nanoemulsification method using VCO and olive oil as an oil phase, Tween 80, Tween 20 and Cremophor RH40 as a surfactant, and polyetilene glykol (PEG) 400 as a cosurfactant. Evaluation of SNEDDS were included of dispersibility test, self emulsification time, globules size, polidispersity index, zeta potential, and stability test by centrifugation and freeze thaw cycles test. Optimal nanoemulsion which contain of 200 PL Nigella sativa oil per I g mixture of oil phase can be made using 10% of VCO, 70% of Cremophor RH 40 and 20% of PEG 400. Dispersibility test showed that this SNEDDS is rapidly form a transparent mixture within 9.17 + 0.015 minutes in the media of HCL 0. IN with the rate of stirage at 50 rpm. This SNEDDS has a mean globules diameter of 57.6 + 13.04 nm, polydispersity index of 0.288 ± 0.033, and zeta potential of 0.243 ± 0.136 mV. Stability test results of centrifuge test showed that SNEDDS of Nigella sativa oil is physically stable, although at the third cycle of freeze thaw cycle showing a significant difference in polydispersity index and zeta potensial with the previous cycle. Result of anti inflammation acute activity test of Nigella sative oil which is incorporate in SNEDDS of VCO has no anti inflammation.
