Assay of NO is usually done indirectly through its main metabolites nitrite and nitrate as an index of NO. NO levels in the body will increase with the presence of inflammation, metabolic syndrome, diabetes, and also plays an important role in the pathogenesis of rheumatoid arthritis. NO acts as reactive intermediates that can induce cellular autoimmunity and contribute to the prolonged innate immune response, as well as in organ damage. NO can be used to predict disease progression and status. Methods: The developed method was determining total nitrite concentration in serum which applied Griess reaction. The reduction of nitrate to nitrite was performed using copper coated zinc. The method was optimized and validated. The determination of total nitrite in samples was started with deproteinization using ZnSO4 and NaOH. Then the supernatants were reducted using copper coated zinc, reacted with diazotization and coupling reagents, then incubated. The absorbance was measured using spectrophotometer visible. Results: The optimum reductant mass of copper coated zinc is 1.5 g with reduction time 10 minutes, coefficient variation ?5%. Linierity of analytical method gives a coefficient correlation, r > 0.995. Relative procedural standard deviation (Vx0) was ?5%. Conclusion: Mean of total nitrite of each sample groups were higher than normal value, and patients are suspected having three to five of metabolic syndrome risk factors. Fasting and post prandial glucose have no significant correlation with total nitrite. There is no significant difference between total nitrite in women and men. While total nitrite in CRP positive and negative significantly different . text
