Summary:Mucoadhesive tablet is a solid dosage form that produced by incorporation of mucoadhesive polymers that expected to attach to gastrointestinal tract mucous to prolong gastrointestinal tract residence time. Combination of this mechanism with the extended release properties are intended to decrease drugs administration frequency. In this study, salbutamol sulfat mucoadhesive extended-release tablet was developed by using mucoadhesive hydrophilic polymers, polyethylene oxide (PEO) and sodium carboxymethylcellulose (CMCNa). Tablets were prepared by direct compression method using varied PEO-CMCNa polymer combination as percentage fraction by total tablet weight (10:0; 10:5; 10:7,5; 20:0; 20:2,5; 20:5). Drug release study was performed using USP type 2 dissolution apparatus in HCl pH 1,2 medium for 12 hour. Then, sample was analyzed using UV spectrophotometric method. Drug release data was extrapolated into four different release kinetic equations (zero order, first order, Higuchi, and Krosmeyer- Peppas). Formula exhibited release properties that met requirements of extended-release criteria was evaluated for the in vitro mucoadhesive properties on rabbit gastric mucous using wash-off method. From the evaluation data, formula exhibited salbutamol sulfat release profile that met requirement contained 10:7,5 and 20:5 PEO-CMCNa combination ratios. Salbutamol sulfat release mechanism from that formula were followed the Higuchi and first order release kinetics. From the in vitro mucoadhesion test in Krebs buffer medium mucoadhesion properties of this formula are 360 and 80 minutes, respectively. To that, the best formula with drug release profile that met requirement and a good mucoadhesive properties was formula with 10:7,5 PEO-CMCNa combination ratio.
