Method: Determination of extraction is carried out using three methods, namely meceration, reflux, and UAE. Then each extract resulting from different extraction methods was tested for flavonoid content, phenol content, and antioxidant activity test. After determining the extraction method, the isolation process was carried out by extraction using UAE, fractionation using vacuum liquid chromatography, and classical column chromatography, and compound identification using 1D and 2D NMR instruments. Results: The highest total flavonoid content was the extract using the UAE method of 7.115 ± 0.226 g QE/100 g, the highest total phenol content was the extract using the UAE method of 10.104 ± 0.345 g GAE /100 g, the antioxidant activity with the lowest IC50 value was amounted to 23.147 ± 0.314 ?g/ml in the very strong category. H-NMR analysis showed that there were 48 H atoms with predicted C-C and C=C. C-NMR analysis obtained results in the form of 30 C atoms with predicted C-C, C=C, and C of carbonil. DEPT analysis resulted in the presence of CH/CH3, CH2, and C quartener molecules in the isolate. HSQC analysis obtained results in the form of C=C and C=O double bonds. HMBC analysis obtained results in the form of neighboring C=O carbonyl bonds with C=C double bonds. Conclusion: The conclusion of this research is that the extraction method that has the best capability is UAE and isolation of secondary metabolite compounds obtained by compound 4,4,9,10,13,14,17-heptamethyl-20- metileneicosahydropicen with chemical formula C30H48O which is a secondary metabolite compound of the triterpenoid group. text |
