The in vitro effects of PGF2a on the caudal epididymis and vas deferens contractility of Wistar rat has been performed. Sexually mature male rats with a body weight of 200-300 g were used, whereas the concentration of PGF2a were 0.4 and 0.6 mg/ml Tyrode. Caudal epididymis and vas deferens were isolated and placed in Tyrode solution at 37 degree C. The contraction of these organs were recorded using a myograph and physiograph. Prior to the PGF2a treatment, the contraction of the organs were recorded for 2.5 minutes, then PGF2a solution was added to the Tyrode solution and the contraction were then recorded for 2.5 minutes hour for the caudal epididymis and every 30 minutes for vas deferens. The observation was stopped when the contraction of the organs reached the normal contraction. The results showed that PGF2a increased the tonus tensions of caudal epididymis as well as vas deferens. Tonus tension at 0.6 mg/ml PGF2a was higher than at 0.4 mg/ml both in the caudal epididymis and vas deferens. At the caudal epididymis, duration of the tonus tension at 0.4 and 0.6 mg/ml PGF2a were 8 and 10 hours respectively, and at the vas deferens were 1.5 and 2 hours. It could be concluded that PGF2a increased the contraction of the caudal epididymis and vas deferens, and acted longer on the caudal epididymis than on vas deferens. |
