Summary:Relapse of addiction is the main problem in tackling nikotin addiction. Arachidonic acid cascade had been found to modulate the relapse of addiction. Therefore, this research aimed to investigate further the role of arachidonic acid cascade in addiction and its relapse. Conditioned Place Preference (CPP) method was used to investigate the addictive effects of nikotin. Addiction was induced by intraperitoneal injection of nikotin at a dose of 0.5 mg/kg. After five days of training, the nikotin injection was ceased for seven days. On the eighth day, animal subject was given a priming injection of nikotin and animal behavior was observed. Animal subject was sacrificed, the hippocampus was isolated, and its protein profile was observed by using SDS-PAGE. After seven days of abstinence, any indication of nikotin addiction seemed to disappear. However, priming of nikotin addiction reinstated the addiction. A diclofenac injection given before the nikotin priming seemed to attenuate the reinstatement of addiction. Analysis of the hippocampus protein profile seemed to show an elevation of arachidonic acid cascade activity during addiction
