Summary:Phaleria macrocarpa and Caesalpinia sappan have been used in traditional cure as antiinflammatory. In vitro assay for isolates of sappan heartwood anti-inflammatory activity has been reported. This study aims to evaluate the anti-inflammatory effect of single extracts, ethanol extract of pericarp of mahkota dewa and methanol extract of sappan heartwood, and combination effect of both extracts. Simplicia used in this experiment are dried pericarp of mahkota dewa and sappan heartwood. Mahkota dewa pericarp and sappan heartwood were extracted using reflux method. Anti-inflammatory assay include observation in redness index, swelling, and walking activity in inflammatory sodium urateinduced rat. Ethanol extract of mahkota dewa pericarp 22,5 mg/kg bb and methanol extract of sappan lignum 63,9 mg/kg bb showed anti-inflammatory effect, but combination of extracts did not perform a better effect. Ethanol extract of mahkota dewa pericarp reduced redness index significantly (p<0.05) on forth and seventh day. Methanol extract of sappan lignum and combination extract reduced redness index significantly (p<0.05) on seventh day. From the third day, combination of extracts reduced walking activity significantly (p<0.05). Single extract reduced walking activity significantly (p<0.05) from forth day. Ethanol extract of mahkota dewa pericarp 22,5 mg/kg bb, methanol extract of sappan lignum 63,9 mg/kg bb and combination of extracts reduced swelling significantly (p<0.05) from the fifth day.
