Summary: | The incidence of Dengue Fever (DBD) tripled in 2024 compared to 2023 in Indonesia. This disease is caused by the dengue virus transmitted by Aedes aegypti and Aedes albopictus mosquitoes. The dengue virus has four serotypes: DENV-1, DENV-2, DENV-3, and DENV-4. The slow detection process often results in severe infections by the time it is detected. Therefore, there is a need for a fast and accurate detection tool. One of the methods developed is the electrochemical biosensor. This research focuses on the development of bimetal MOF CuCoBTC material, including CuBTC, CuCoBTC 1:1, CuCoBTC 1:2, CuCoBTC 1:5, and CuCoBTC 1:10, modified with TEOA as an electrochemical biosensor for detecting the NS1 antigen of dengue virus serotype 3. The research methods include material synthesis using the co-precipitation method, characterization using SEM, XRD, FTIR, and SPA, as well as electrochemical testing using CV, DPV, and EIS techniques.
The research results show that CuCoBTC 1:1 material has the best performance as an electrochemical biosensor. Electrochemical testing indicates that this material has the highest electrochemical response compared to other material variations. Characterization shows that CuCoBTC 1:1 material has a high surface area, which enhances electrochemical performance. Analytic tests demonstrate a significant current reduction when antibodies, BSA, and antigens are added, indicating specific interactions with bioanalytes. The determination of the Limit of Detection (LoD) shows that this biosensor has an LoD of 0.555 pg/ml, better than previous studies.
