An????oxidants are a group of compounds that counteract free radicals within cells, providing protec????on against cell damage caused by free radicals. Kratom ( Mitragyna speciosa ) is a shrub that grows abundantly in Kalimantan and its leaves are tradi????onally consumed as tea to boost stamina. This study aims to determine the compounds with an????oxidant ac????vity in kratom leaves using the DPPH method. The results showed that kratom leaf extracts with ethyl acetate and 96% ethanol solvents had very high an????oxidant ac????vity with IC 50 values of 37.73±2.09 µg/ml and 15.89±0.32 µg/ml, respec????vely. The ethanol extract was frac????onated using vacuum liquid chromatography (VLC). Frac????ons 2, 3, and 8 qualita????vely showed an????oxidant ac????vity and were therefore subjected to purifica????on. Purifica????on with radial chromatography (RC) yielded 4 pure isolates (F2.1, F2.2, F3, F8). Isolate F2.1, which belongs to the alkaloid group, demonstrated an inhibi????on percentage of 15.82 ± 6.26 at 500 µg/ml; F2.2, which belongs to the steroid/triterpenoid group, had an IC 50 value of 37.50±0.84 µg/ml; predicted to be a phenylpropanoid compound had an IC 50 value of 170.37±0.25 µg/ml; and F8, which is predicted to be a terpenoid glycoside subgroup sesquiterpenoid had an inhibi????on percentage of 18.16±2.72 at 100 µg/ml. From the experimental results, it can be concluded that the 96% ethanol extract had the highest an????oxidant ac????vity in the stepwise extrac????on process, with an IC 50 value of 15.89 ± 0.32 µg/ml. The isola????on yielded ac????ve an????oxidant isolates from the alkaloid group (F2.1), steroid/triterpenoid group (F2.2), phenylpropanoid group (F3), and sesquiterpenoid group (F8). text
