One of the ways to solve this problem is by using permeation enhancers (PE). Previous studies have shown that gliadin-derived peptides have potential application as PE. CPP have been shown to display PE activity that can be utilized to improve drug absorption and bioavailability by enhancing the delivery of drugs across bio-membranes. Based on a prior in silico study, p177-195 is an ????-gliadin-derived peptide which has been predicted to be a potential cell penetrating peptide (CPP) with a PE activity. Hence, this study is done to evaluate p177-195 potential in enhancing the permeation of FITC-Dextran 10 kDa (FD10), FITC-Dextran 40 kDa (FD40), and insulin through intestinal epithelial barrier in vivo with Caco-2 monolayer cell as model. p177-195 peptide with a concentration up to 200 ?M doesn’t have any cytotoxic effect on Caco-2 cells. The presence of p177-195 with the concentration of 5 – 150 ?M enhance Caco-2 monolayer cell permeability towards FD10 until 19.90%, FD40 until 11.86%, and insulin. Based on this study, p77-195 is a candidate for potential CPP to be furtherly studied safety and efficacy as a permeation enhancer. text |
