According to several studies, natural materials with abundant availability, namely eggshell membrane (ESM), have been identified to have potential in wound healing. Although ESM can accelerate wound healing, its solid form cannot penetrate the skin, necessitating its use in soluble hydrolysate form. Enzymatic hydrolysis of ESM is conducted using the papain enzyme, with the addition of sodium sulfite. This study aims to evaluate the wound healing activity of hydrolyzed ESM (HESM) fractions. HESM was fractionated into Fraction I (<10 kDa), Fraction II (10-30 kDa), and Fraction III (>30 kDa) using ultrafiltration membranes. Each HESM fraction was tested for antioxidant activity using DPPH and its wound healing ability through an in vitro wound healing scratch assay. The antioxidant activity of Fraction II was higher compared to other HESM fractions. Cell viability tests showed that Fraction I, Fraction II, and Fraction III at concentrations of 500 to 1500 ppm were non-toxic. The highest wound closure percentage within 24 hours was achieved by Fraction I at a concentration of 250 ppm, indicating that Fraction I has the best capability to induce 3T3 cell migration and proliferation, thereby accelerating the wound healing process. text |
