Karamunting leaves (Rhodomyrtus tomentosa (Aiton) Hassk.) is one of plants used in traditional medication for various health conditions, including inflammation. Therefore, this study aims to find out the antiinflammatory activity of karamunting leaves extracts and fractions using protein denaturation, blood membrane stabilization methods, and find out the active fraction compounds characteristics contained. The samples were tested for antiinflammatory activity in vitro using BSA protein denaturation and blood membrane stabilization. Selected fractions were characterized by TLC monitoring and TLC scanner. It was found that the antiinflammatory activity ethanol extract of karamunting leaves (Rhodomyrtus tomentosa (Aiton) Hassk.) has inhibitory activity against BSA protein denaturation and hemolysis inhibition on red blood cells with respective IC50 values consecutively in 149,238 ± 5,832 and 243,775 ± 5,617 mg/mL. The IC50 values of fraction 1, fraction 2, fraction 3, fraction 4, and fraction 5 are respectively 1000,970 ± 25,941; 604,615 ± 82,045; 497,797 ± 5,102; 162,992 ± 0,594; 105,203 ± 10,862 mg/mL. Fraction 5 is a fraction that has the best denaturation inhibition activity value characterized by the smallest IC50 value. The compound groups contained in fraction 5 are flavonoids, phenols, tannins, and saponins. The compound successfully identified using the KLT densitometry method was the eugenol compound. text
