Various experiments have been conducted to find new drug candidates to solve the antimicrobial resistance problem, one of the solutions is by looking for secondary metabolite compounds from marine microbes. Aspergillus flocculosus (code FSB-27) is a fungi collection from the Microbiology Laboratory, School of Pharmacy ITB isolated from a coral in Tulamben, Bali. Aspergillus flocculosus shows antimicrobial activity against Candida albicans, Escherichia coli, Staphylococcus aureus, and Mycobacterium smegmatis. This study was aimed to obtain a fraction of secondary metabolites of Aspergillus flocculosus that have antimicrobial activity. This research includes the rejuvenation process, identification, fermentation, extraction of secondary metabolites, fractionation of secondary metabolites, subfractionation of secondary metabolites, and activity tests on microbes. Activity test using disc diffusion is used in antibacterial and antifungal activity screening and microdilution is used in determine the minimum inhibitory concentration (MIC). In antimycobacterial, the MTT test method is used for activity screening and determination of the MIC value. The minimum bactericidal/fungicidal concentration value (MBC/MFC) is conducted by drop test. The results showed media extract had activity against all test microbes, while the mycelium extract gave activity to Staphylococcus aureus. Methanol and hexane fraction had activity against all test microbes. The results of the subfraction from methanol fraction showed that subfraction D gave moderate activity against Escherichia coli, Staphylococcus aureus, and Mycobacterium smegmatis with MIC respectively 500, 250, and 500 ?g/mL. Subfraction D gave moderate fungicidal activity against Candida albicans with MFC 250 ?g/mL. Subfraction C and E gave weak activity against Candida albicans with MIC 1000 ?g/mL. In general, the fractionation results of ethyl acetate extract of Aspergillus flocculosus culture is a potential source of antimicrobials. text
