Summary:Jati belanda leaves (Guazuma ulmifolia L.) are known to contain tiliroside in their extract, which has the biological activity of reducing cholesterol levels in the blood. Seeing its biological activity, jati belanda leaves can be used as an alternative treatment for dyslipidemia. In monitoring the quality of jati belanda leaves extracts and nano extracts, a validated analytical method is needed to determine tiliroside levels. This research aims to develop and validate analytical method for determining tiliroside levels in jati belanda leaves extracts and nano extracts using High Performance Liquid Chromatography (HPLC). Based on the optimization results, the chromatography system used was a stationary phase of the Agilent ZORBAX Eclipse XDB-C18 equipped with a guard column, a mobile phase of 100% acetonitrile and 0.1% formic acid in 10% acetonitrile was used with gradient elution method, injection volume 10 ?L, flow rate 1 mL/min, and the detection wavelength is 330 nm. Under optimum conditions, The analytical method developed has met the criteria and proven to have good robustness. The calibration curve shows a linear relationship with an r = 0.999 and Vxo = 1.669% in the concentration range of 1.8 – 14.4 ?g/mL. The detection and quantitation limits were obtained 0.482 ?g/mL and 1.460 ?g/mL. In testing methods precision and interday precision, RSD values were obtained of 1.496% and 3.622% for extracts, 1.265% and 1.651% for nano extracts. The average value of recovery in accuracy measurements is in the range of 99.535 – 101.604 % for extracts and 97.123 – 98.323 % for the nano extracts. Determination of tiliroside levels using a validated method showed that tiliroside levels in the extracts and nano extracts jati belanda leaves samples were 0.127 ? 0.002 % and 0.294 ? 0.001 %.
