Denatured proteins result in reduced enzymatic activity and biological function of proteins, and can trigger the production of autoantigens in the body. Compounds that can inhibit protein denaturation have potential as antiinflammatory agents. Cocor bebek (Kalanchoe pinnata (Lam.) Pers.) is a plant that can be developed as an alternative antiinflammatory treatment by inhibiting protein denaturation due to its rich content of secondary metabolites, such as flavonoid, phenol, and steroid/triterpenoid compounds. The study was conducted to determine the profile of chemical compounds in the leaf extract of cocor bebek and identify the antiinflammatory activity of the extract by inhibiting protein denaturation. Extraction was performed using a maceration method with n-hexane, ethyl acetate, 96% ethanol, and 50% ethanol as solvents. Chemical compound profiles were determined based on phytochemical screening, determination of total phenol and flavonoid content, and analysis using TLC densitometry. Antiinflammatory activity was determined using UV-Visible spectrophotometer at ? 660 nm, with sodium diclofenac as a positive control. Phytochemical screening showed the presence of flavonoid, phenol, and steroid/triterpenoid compounds in cocor bebek leaf extracts. The 96% ethanol extract had the highest total phenol content (5.58 ± 0.37 g GAE/100 g), while the ethyl acetate extract had the highest total flavonoid content (4.78 ± 0.29 g QE/100 g). The IC50 values of n-hexane, ethyl acetate, 96% ethanol, and 50% ethanol extracts of cocor bebek leaves were 385.91 ± 2.52 ppm, 143.39 ± 1.47 ppm, 920.52 ± 6.06 ppm, and 1295.64 ± 5.76 ppm, respectively. The flavonoid compounds identified in the ethyl acetate extract were quercetin-3-arabinoside and luteolin with levels of 17.15% and 30.62%, respectively. Cocor bebek leaf extract has moderate potential to be developed as an alternative antiinflammatory treatment. text
